Abstract:

The fault of wheelset bearing is one of the most important factors which is deangerous to the satfy of the train running. And the key problem that should be solved firstly is the accur ate measurement method for the high speed heavy haul locomotive. Since there are some discontinuity points in the amplitude of the vibration signals when the wheelset bearing is abnormal, a method for analyzing the vibration acceleration signals which come from the wheelset bearing by using the spectrum zoom is prsented. The experiments indicate that the abnormity of wheelset bearing high-accuracy detection can be realized by using this method, and this method can detect the abnormity effectively compared to the commonly used methods.

Feature extraction of hyperspectral images for detecting immature green citrus fruit

Abstract:

At an early immature growth stage of citrus, a hyperspectral camera of 369–1042 nm was employed to acquire 30 hyperspectral images in order to detect immature green fruit within citrus trees under natural illumination conditions. First, successive projections algorithm (SPA) were implemented to select 677, 804, 563, 962, and 405 nm wavebands and to construct multispectral images from the original hyperspectral images for further processing. Then, histogram threshold segmentation using NDVI of 804 and 677 nm was implemented to remove image backgrounds. Three slope parameters, calculated from the pairs 405 and 563 nm, 563 and 677 nm, and 804 and 962 nm were used to construct a classifier to identify the potential citrus fruit. Then, a marker-controlled watershed segmentation based on wavelet transform was applied to obtain potential fruit areas. Finally, a green fruit detection model was constructed according to Grey Level Co-occurrence Matrix (GLCM) texture features of the independent areas. Three supervised classifiers, logistic regression, random forest and support vector machine (SVM) were developed using texture features. The detection accuracies were 79%, 75%, and 86% for the logistic regression, random forest, and SVM models, respectively. The developed algorithm showed a great potential for identifying immature green citrus for an early yield estimation.

Abstract: H2S is well-known as a colorless, acidic gas, with a notoriously rotten-egg smell. It was recently revealed that H2S is also an endogenous signaling molecule that has important biological functions, however, most of its physiology and pathology remains elusive. Therefore, the enthusiasm for H2S research remains. Fluorescence imaging technology is an important tool for H2S biology research. The development of fluorescence imaging technology has realized the study of H2S in subcellular organelles, facilitated by the development of fluorescent probes. The probes reviewed in this paper were categorized according to their chemical mechanism of sensing and were divided into three groups: H2S reducibility-based probes, H2S nucleophilicity-based probes, and metal sulfide precipitation-based probes. The structure of the probes, their sensing mechanism, and imaging results have been discussed in detail. Moreover, we also introduced some probes for hydrogen polysulfides.

A dual-function chemical probe for detecting erasers of lysine lipoylation

Abstract: Lysine lipoylation plays vital roles in cell metabolism and redox processes. For example, removal of lipoylation will decrease pyruvate dehydrogenase activity and affect the citric acid cycle. Despite the important functions of lysine lipoylation, the mechanisms for the addition and removal of this modification remain largely unexplored. Very few useful chemical tools are available to study the interactions of lysine lipoylation with its regulatory delipoylation proteins. For example, immunoaffinity purification-mass spectrometry is one of such tools, which highly relies on antibody efficiency and purification techniques. Single-step activity based fluorogenic probes developed by our groups and others is also an efficient method to study the deacylation activity. Affinity-based labeling probe using photo-cross-linker is a powerful platform to study the transient and dynamic interactions of peptide ligands with the interacting proteins. Herein, we have designed and synthesized a dual-function probe KTLlip for studying enzymatic delipoylation (eraser) activity and interaction of lysine lipoylation with the eraser at the same time. We show that KTLlip can be used as a useful tool to detect delipoylation as demonstrated by its ability to fluorescently label the eraser activity of recombinant Sirt2. We envision that the probe will help delineate the roles of delipoylation enzyme in biology.

Abstract: nitrosodimethylamine (NDMA) and several other nitrosamines have been detected as disinfection by-products in drinking waters in many countries around the world. An ultra-performance liquid chromatography-tandem mass spectrometry method with solid phase extraction sample preparation was developed to study the occurrence of nitrosamines in several water treatment plants and distribution systems in China. Isotope labeled nitrosodi- propylamine-d14 (NDPA-d14) was selected as the internal standard for quantification. The solid phase extraction procedures including pH, enrichment process and MS/MS parameters including capillary voltage, cone gas flow, cone voltage, collision energy were optimized to give average recoveries of 26% to 112% for nine nitrosamine species. The instrument detection limits were estimated to range from 0.5 to 5 μg·L for the nine nitrosamine species. NDMA and several other nitrosamines were found at fairly high concentrations in several water treatment plants and distribution systems. NDMA was found in all locations, and the highest concentrations in cities B, G, T, and W were 3.0, 35.7, 21.3, and 19.7 ng·L , respectively. A wide range of nitrosamines concentrations and species were observed in different locations. Higher concentrations of nitrosamines were detected in distribution systems that were further away from the treatment plants, suggesting that the contact time between the residual disinfectant and natural organic matter may play an important role in the formation of these compounds.
